While normal plasma cells produce antibodies, in myeloma patients they build-up abnormally and release only one type of antibody, called paraprotein, which has no useful function.

There are treatments for symptoms and complications of myeloma patients; however, since scientists have not found a cure yet, nearly all patients eventually die from the disease. The good news is that having found an underlying cause of multiple myeloma, which was a mystery until now, could help to open new lines of research for a better treatments.

The study, published Feb. 10 in the New England Journal of Medicine, showed that a third of all myeloma cases are originated by chronic stimulation of the immune system by lipids made in the context of inflammation.

“Understanding the origin of any cancer has several implications for how to best prevent it,” said Senior author Dr. Madhav Dhodapkar. “These studies set the stage for newer approaches to lower the levels of these lipids in patients with Gaucher disease and others with precursors for myeloma. Potentially, this could be achieved with drugs or lifestyle changes to reduce the levels of lipids to lower the risk of cancer.”

The new discovery arose from Dhodapkar’s previous research on Gaucher disease, in which he found that the patients with this inherited lipid storage disorder, have a significant increased risk for developing myeloma. He also discovered a subgroup of lipid-reactive immune cells that promote the development of plasma cells, called type II NKT-TFH.

In the current research, using tissue and blood samples of mice models of Gaucher disease and humans with Gaucher disease, the researchers found that gammopathy (a precursor to myeloma) is triggered by specific lipids, lysolipids LGL1 and lysophosphatidylcholine (LPC). Furthermore, it was discovered that the antibodies released by the cancerous plasma cells in nearly a third of myeloma patients are directed against such lipids.

Source: Yale News

Yale researchers found a mechanism that causes a third of all cases of myeloma, a type of cancer in which plasma cells become abnormal and multiply uncontrollably.
