The finding reveals a new possible target to treat ovarian and other metastatic cancers.

Ovarian Cancer is the most aggressive gynecological malignancy and a leading cause of death. According with the article, published in the Journal of Clinical Investigation, its high mortality is due to its difficult diagnosis in early stages, which is only commonly achieved until stage III — when it has spread beyond the pelvis to the lining of the abdomen, and/or has spread to lymph nodes in the back of the abdomen.

This intraperitoneal and extensive pelvic implantation metastasis is the most important cause of poor prognosis of ovarian cancer, as it is usually unable to be removed completely by surgery. During metastasis, ovarian cancer cells detach from the tumor and migrate to the abdominal cavity, where their survival is supported by tumor-associated macrophages (TAMs).

The study led by Wang Min, Yale pathology professor, showed that tumors release substances called cytokines to attract macrophages, which then secrete growth factors that promote tumor cell growth and proliferation.

The study specifically demonstrated that TAMs play an essential role in spheroid formation during ovarian cancer metastasis. TAM can secrete large amounts of epidermal growth factor (EGF) to activate its receptor (EGFR) in surrounding tumor cells. The activated EGF/EGFR signaling can upregulate vascular endothelial growth factor C (VEGF-C), which overexpression is linked with several cancers.

This in turn upregulates integrins and a protein called ICAM-1, to form a positive autocrine feedback loop, thus promoting tumor cell proliferation, migration, adhesion, spheroid formation, and peritoneal implantation .

“Our current study has unveiled the underlying mechanism of spheroid formation, providing a strategy for inhibiting implantation metastasis and improving the prognosis of OC patients.”

Source: Yale News, Journal of Clinical Investigation

Yale researchers found a mechanism used by tumors to promote its growth and proliferation.
