This issue has caused that sometimes this condition is known as ‘the invisible pain’ and that those who suffer it feel misunderstood, presenting a state of permanent frustration that often aggravates their condition.

Fibromyalgia statistically affects women more than men. Its causes are unknown and are associated with different events that include viral infections, surgical operations, births, affective events and even abuses.

The new study included a cohort of 28 women diagnosed with fibromyalgia according to the criteria of the American College of Rheumatology aged between 28 and 55 years; who were compared with a control cohort of healthy volunteers, ages between 28 and 51.

The work provides a small-scale genetic signature composed of 57 genes for diagnosis. The analyzes showed that the three major genetic pathways affected are related to the activation of hepatic stellate cells (liver adipocytes, indicative of injuries in this organ), oxidative phosphorylation, and respiratory diseases such chronic obstructive pulmonary disease (COPD).

Although the results are preliminary and difficult to interpret, it seems to confirm an important role of the signaling pathway of glutamate, a neurotransmitter with great importance in chronic fatigue and depression. They also present an altered expression of genes related to the attenuation of the metabolic rate and of the inflammatory pathways associated with Interferon alfa, a cytokine involved in the regulation of the immune response against infections.

Searching new therapeutic targets

Juan Luis Fernández-Martínez, an expert from the University of Oviedo, emphasizes the importance of translational research which results impact on the treatment patients receive.

“In the absence of clinical confirmation, the findings connect fibromyalgia with chronic fatigue syndrome. In the future we would like to study the differences between them, as well as the mechanisms that are common and compare them with the altered pathways in multiple sclerosis,” explains Fernández-Martínez.

The teams that participated in this work have also been collaborating for some time studying the mechanisms involved in the development of chronic fatigue in cancer patients treated with radiotherapy.

These results not only have a diagnostic value, but also allow searching new therapeutic targets and the using drugs that are able to optimally regulate the altered genes and minimize the side effects.

Source: SINC

Scientists at the University of Oviedo, in collaboration with experts from Johns Hopkins University in Baltimore and the National Institute of Nursing Research at the National Institute of Health (NINR-NIH), described the major genetic pathways altered in patients with fibromyalgia.
